Colorectal cancer combination therapy using drug and gene co-delivered, targeted poly(ethylene glycol)-ε-poly(caprolactone) nanocarriers

摘要: Purpose Combination therapy is a promising strategy to treat cancer due the synergistic effects. The drug and gene co-delivered systems attract more attention in field of combination therapy. Materials methods In present research, poly(ethylene glycol)-e-poly(caprolactone) block copolymer was used for co-loading 5-fluorouracil (5-FU) gene. physicochemical characteristics, vitro vivo anticancer, transfection efficiency were tested on colon cells tumor-bearing mice. Results 5-FU co-loaded nanocarriers had size 145 nm. delivery results showed about 60% gene-positive cells. Tumor volume nanocarrier groups at day 21 around 320 mm3, which significantly smaller compared with free group (852 mm3) control (1,059 mm3). maximum plasma concentration (49 µg/mL) greater than (13 µg/mL). At 24 hours, level 2.8 µg/mL 0.02 5-FU. Conclusion resulting anticancer drugs genes could be considered as nanomedicine colorectal
