摘要: Introduction: Inflammatory mediators are key players in the pathogenesis of osteoarthritis (OA) and bone destruction. Conventional drugs suppress symptomatic activity have no therapeutic influence on disease. Cissus quadrangularis Withania somnifera widely used for treatment fractures wounds; however, cellular molecular mechanisms regulated by these herbals still unclear. Methods: We established an vitro OA culture model exposing human chondrocytes to proinflammatory cytokine interleukin (IL)-1 β 36 hours prior with herbals: C. quadrangularis, W. somnifera, combination two herbals. Cell viability, toxicity, gene expression modifying agents were examined. In addition, survivin, which is crucial cell growth, was analyzed. vivo work osteotomized rats studied cartilage regenerative effects therapy. Results: Exposure IL-1β induced significant toxicity death. However, herbal alleviated upregulated growth proliferation. inhibited cytokines matrix metalloproteinases, known aggravate destruction, augmented survivin inhibiting p38 MAPK. Interestingly, treated drastically enhanced alkaline phosphatase tissue formation as compared untreated, only, or both Conclusion: Our findings demonstrate first time signaling osteogenesis. suggest that chondroprotective ability via upregulation exerts inhibitory MAPK pathway. These thus validate
