Correlation of murine embryonic stem cell gene expression profiles with functional measures of pluripotency.

摘要: Global gene expression profiling was performed on murine embryonic stem cells (ESCs) induced to differentiate by removal of leukemia inhibitory factor (LIF) identify genes whose change in correlates with loss pluripotency. To appropriate time points for the analysis, dynamics pluripotency were investigated using three functional assays: chimeric mouse formation, embryoid body generation, and colony-forming ability. A rapid detected within 24 hours, very low residual activity all assays 72 hours. Gene profiles undifferentiated ESCs cultured 18 hours absence LIF determined Affymetrix GeneChip U74v2. In total, 473 identified as significantly differentially expressed, approximately one third having unknown biological function. Among 275 decreased ESC differentiation several factors previously important for, or markers of, pluripotency, including Stat3, Rex1, Sox2, Gbx2, Bmp4. significant number also overlap published human data. Furthermore, membrane proteins among 48 correlating most closely assays, cell receptor c-Kit. Through identification follows properties during early differentiation, this study lays foundation further elucidating molecular mechanisms regulating maintenance facilitates more reliable state.
