摘要: A radioimmunoassay specific for liver pyruvate kinase was used to determine the mechanism(s) involved in insulin stimulation of this enzyme activity chronically diabetic rats. Rats, made with alloxan, were fed on a high-carbohydrate (50%-sucrose) fat-free diet and treated 12, 36 or 60 h. Livers removed at various times, piece kept determination glycogen, remainder homogenized. The 100000 g supernatant prepared quantity. Glycogen increased maximum approx. 7% by 12 h after treatment, maintained elevated value Liver activity, which is depressed diabetes, did not respond until more substantial increase occurring Radioimmunoassay data indicated that concomitant quantity moderate its activity. These results demonstrate dual mechanism, i.e. an both enzyme, regulates insulin-mediated rat.
