摘要: Yeast Saccharomyces cerevisiae strains that are permeable to the antitumor alkaloid camptothecin killed by drug if they express DNA topoisomerase I, cellular target of (J. Nitiss and J.C. Wang, Proc. Natl. Acad. Sci. USA, 85: 7501-7505, 1988). We show in a yeast strain but lacking sensitivity was restored upon expression human I from plasmid-borne complementary clone. When enzyme expressed galactose-inducible, glucose-repressible promoter, PGAL1, observed presence galactose not glucose. Expression Escherichia coli also demonstrated complementation conditional lethal E. mutant. These systems can be used study I-camptothecin interactions screening additional therapeutics targeting enzyme.
