摘要: Quantitative autoradiography was used to determine the distribution of L-[3H]glutamate-binding sites in rat central nervous system. Autoradiography carried out presence Cl- and Ca2+ ions. Scatchard plots Hill coefficients glutamate binding suggested that interacting with a single population having K-D about 300 nM capacity 14.5 pmol/mg protein. In displacement studies, ibotenate also appeared bind class non-interacting KI 28 microM. However, quisqualate [3H]glutamate revealed two well-resolved KIS 12 114 microM striatum. These were unevenly distributed, representing different proportions specific brain regions. The glutamate- correlated very well projection areas putative glutamatergic pathways. This technique provides an extremely sensitive assay which can be gather detailed pharmacological anatomical information L-[3H]glutamate
