摘要: Recovery of cell volume in response to osmotic stress is mediated part by increases the Cl- permeability plasma membrane. These studies evaluate hypothesis that ATP release and autocrine stimulation purinergic (P2) receptors couple opening channels. In HTC rat hepatoma cells, swelling induced hypotonic exposure increased membrane current density 44.8 +/- 7.1 pA/pF at -80 mV. Both rate recovery increase were inhibited presence hydrolase apyrase (3 units/ml) or P2 receptor blockers suramin Reactive Blue 2 (10-100 microM). Cell also stimulated ATP. Hypotonic concentration effluent perfused cells 170 36 nM a nucleotidase inhibitor (P < 0.01). whole-cell recordings with as charge carrier, approximately 30-fold 16.5 10.4 pA/pF. findings indicate lead efflux through conductive pathway consistent channel, extracellular required for from swelling. may function an factor couples channels receptors.
