摘要: Allergenic proteins such as grass pollen and house dust mite (HDM) are known to trigger hypersensitivity reactions of the immune system, leading what is commonly allergy. Key allergenic including sequence variants have been identified but characterization their post-translational modifications (PTMs) still limited. Here, we present a detailed PTM(1) series main clinically relevant allergens used in allergy tests vaccines. We employ Orbitrap-based mass spectrometry with complementary fragmentation techniques (HCD/ETD) for site-specific PTM by bottom-up analysis. In addition, top-down utilized targeted analysis individual proteins, revealing hitherto unknown PTMs HDM allergens. demonstrate presence lysine-linked polyhexose glycans asparagine-linked N-acetylhexosamine on Moreover, more complex glycan structures than previously reported major group 1 5 allergens, implicating important roles carbohydrates allergen recognition response system.
