摘要: This work is a comparative study on the features displayed by an epithelial metastatic breast cancer cell line (MCF-7) when set in co-culture with human bone marrow mesenchymal stem cells (MSC) or feeder layer of 3T3 fibroblasts. MSC, subset non-haematopoietic stroma, display potential for self-renewal, proliferation and differentiation into precursors bone, cartilage, connective muscular tissue. Adhesion MCF-7 to monolayers MSC was high (95 85% respectively). Once attached, grow well both monolayers. Morphology cells, as analysed light epifluorescence microscopy, revealed that clusters 3T3, but disperse MSC. Concomitant lost their aggregation status, express low levels intercellular adhesion molecules, E-cadherin epithelial-specific antigen (ESA). These results suggest represent appropriate target investigate cellular molecular events occurring at interface epithelial-marrow stromal interactions. Together, model here described should permit further evaluate significance prognostic impact shift micrometastatic from cluster-aggregated single-cell status. © 2000 Cancer Research Campaign
