A novel gene, GCRG224, is differentially expressed in human gastric mucosa

摘要: AIM: To clone genes that may predispose us to human gastric cancer and analyze it’s expression in tissues. METHODS: Specimens of paired tumor, paratumor normal mucosa tissues collected from fifteen patients who suffered stomach antrum adenocarcinoma were used for analysis. Seven out the cases first studied by fluorescent differential display reverse transcription polymerase chain reaction (DDTR-PCR) The differentially expressed bands interest cloned, analyzed Northern blot, sequencing RT-PCR. Through BLAST, results compared with GenBank database homology In situ hybridization DIG-labeled cRNA probes was interesting cDNA paraffin embedded isolated 30 patients. RESULTS: DDRT-PCR showed one bands, which named W2, much higher all seven tested tumor samples than their counterparts, it sub-cloned into a pGEM-T Easy vector. Two subclones subsequently obtained. One subclone, GCRG224, further. result GCRG224 consisted 1159 base pairs had open reading frame (ORF). It located at chromosome 11q14. No homologue found GCRG224-ORF. However, blot analysis tissue one. Human multiple revealed also colon tissue, peripheral blood leukocyte. only 5/30 adenocarcinoma, 3/18 dysplasia 6/18 intestinal metaplasia level glands. over-expressed predominantly 26/30 mucosal epithelium. CONCLUSION: A novel gene identified tissue. overexpressed almost epithelium but small portion precancerous leisions. role needs further study.
