摘要: Cell-free replication systems for simian virus 40 (SV40) DNA are taken to be a model the of eukaryotic chromosomes, because only one viral protein is required supplement proteins provided by human cell extract. To prove that these cellular function in chromosomal we have begun identify homologous an organism can genetically manipulated. Here report identification yeast factor-A (yRF-A) from Saccharomyces cerevisiae and show it functionally structurally related initiation elongation SV40 replication. Yeast RF-A, multi-subunit phosphoprotein, similar its chromatographic behaviour, subunit structure DNA-binding activity. The will fully substitute early stage Substitution yRF-A complete system, however, results reduced replication, presumably due requirement species-specific interactions between RF-A polymerase complex.
