摘要: To determine if epithelial lipoxygenases are regulated by mucosal inflammation, we examined the distribution of arachidonate 12-lipoxygenase in healthy colonic tissue and involved uninvolved sections colon with inflammatory bowel disease. Immunohistochemistry formaldehyde-fixed, paraffin-embedded intestinal using two anti-12-lipoxygenase antibodies indirect biotin-avidin-peroxidase detection demonstrated that, contrast to from normal (n = 8), which antigen was undetectable cells, glandular epithelium inflamed ulcerative colitis 4) or Crohn's disease exhibited markedly positive immunostaining antibodies. Immunoblotting whole cell extracts showed immunoperoxidase staining a single protein band that comigrated purified (in relative molecular weight: M(r) 72,000) samples subjects but no comparable same colons without Assays activity indicated corresponding increase enzymatic samples. The increased levels cells regions affected disease, increases activity, absence detectable types noninflamed all suggest is induced local mediators inflammation.
