摘要: Oligonucleotide-polymer conjugates have been described to improve the sensitivity of an enzyme-linked oligosorbent assay diagnostic test. To understand influence their structure and conformation in solution on efficiency test during capture step, two different ways synthesizing these were compared. The first consisted coupling 5' amino modified oligonucleotides poly(maleic anhydride-alt-methylvinyl ether) anhydride-alt-ethylene). second resulted from direct synthesis anhydride-alt-ethylene) previously grafted onto a controlled pore glass support. analyzed by size-exclusion chromatography viscometry. former method for conjugate produced aggregates, which was not case latter. These then used phase hybridization using HBV DNA target, bioMerieux VIDAS instrument. Different parameters studied, such as purity number per polymer chain. amount coated solid-phase receptacle surface at time evaluated radioactive labeling. Finally, it demonstrated that amplification factor 50 versus oligonucleotide probe reference. detection limit reached 10(8) copies/mL.
