摘要: Purpose : To compare immunohistochemical staining using different anti-p53 antibodies, and to evaluate the possible clinical implications of overexpression p53 in a series patients resected for colorectal cancer with long follow-up. Methods Tumor biopsy samples were collected from 294 surgical specimens, obtained two median follow-up 4.5 years. The stained four commercially available (mouse monoclonal antibodies 421, 1801, DO-7 ; rabbit polyclonal antibody CMI), evaluated compared cryosections subset 20 biopsies tumors various stages grades, outcomes. Results DO- 7 gave homogeneous nuclear staining, which, when further investigated, turned out be identical formalin-fixed paraffin-embedded frozen specimens. Therefore, was found suitable analysis archival or sections sample. shown 162 (55%) cases, significantly higher proportion having DNA aneuploidy (p<0.01) left-sided colonic rectal (p<0.01). not associated tumor stage, grade, survival. Conclusions A overexpressing are aneuploid, indicating that mutations TP53 gene constitute sign genetic instability, which might importance malignant transformation. However, we could find any indication mutations, as reflected p53, prerequisite progression cancer.
