Telomerase-transduced osteoarthritic fibroblast-like synoviocyte cell line

摘要: To examine whether the life span of fibroblast-like synoviocytes (FLSs) can be extended and to establish FLS cell lines that preserve characteristics primary FLSs, we introduced human catalytic subunit telomerase (hTERT) gene into osteoarthritic (OA) FLSs. Two hTERT-transduced clonal were established one line, hTERT-OA 13A, was characterized. The 13A cells have a morphology similar parental untransduced population-doubling time early passages. While OA FLSs reached senescence after 100 days in culture, continued grow at rate once about every 2-3 days. so far grown culture beyond 450 maintained same growth rate. Furthermore, preserved their sensitivity response treatment with basic calcium phosphate crystals interleukin-1beta. In conclusion, exogenous expression represents way extend telomerase-transduced offer promising tool for regulation, cell-based assay, transplantation-based therapy, tissue engineering research development.
