Pyruvate protects against experimental stroke via an anti-inflammatory mechanism

摘要: Pyruvate, a key intermediate in glucose metabolism, was explored as potential treatment models of experimental stroke and inflammation. Pyruvate administered to rodents after the onset middle cerebral artery occlusion (MCAO). Since extent inflammation is often proportional size infarct, we also studied group animals given lipopolysaccharide (LPS) cause brain without cell death. Following MCAO, pyruvate did not affect physiological parameters but significantly reduced infarct volume, improved behavioral tests numbers neutrophils, microglial NFκB activation. Animals LPS showed increased activation which almost completely abolished by pyruvate. Lactate, major metabolite pyruvate, administration. However, administration lactate itself have any anti-inflammatory effects. protects against ischemia possibly blocking inflammation, does appear explain pyruvate's properties.
