摘要: An investigation of virus-specific protein synthesis in infectious pancreatic necrosis virus (IPNV)-infected rainbow trout gonad cells was undertaken to find a relationship between the coding capacity genome (two segments double-stranded RNA 2.5 × 10 6 and 2.3 molecular weight) sizes relative amounts proteins. Using polyacrylamide slabgel electrophoresis autoradiography, eight distinct polypeptides were detected infected, [ 35 S]methionine-labeled cells. These proteins may be grouped into three size classes on basis weight: (i) large, α (90,000); (ii) medium, β 1 (59,000), 2 (58,000), 3 (57,000); (iii) small, γ (29,000), 1A (28,000), (27,000), (25,000). The combined weight these polypetides (373,000) is beyond genome. Purified IPNV contained α, , . Pulse-chase experiments tryptic peptide map comparisons revealed that only four intracellular primary gene products, namely, with 205,000. Of products polypeptide found stable, whereas other underwent proteolytic cleavage during morphogenesis. Polypeptide cleaved generate ; trimmed produce nonstructural product, precursor results suggest possesses unique mechanism synthesize using mRNA transcripts from two high-molecular-weight segments.
