摘要: PURPOSE. CD40 antigen is a membrane receptor that plays role in the regulation of immune reactions. The expressions and ligand (CD40L) were investigated ex vivo vitro conjunctival epithelial cells, correlation with HLA DR class II antigen, previously shown to be upregulated inflammatory conditions. METHODS. Impression cytology specimens collected 186 patients: 52 normal ones, 65 keratoconjunctivitis sicca, 69 chronic conjunctivitis. Cells processed for flow cytometry, by using monoclonal antibodies CD40, CD40L, antigens. Chang cells also used treated human recombinant interferon (IFN)-g or tumor necrosis factor (TNF)-a. studied cytometry after 24 48 hours treatment. RESULTS. was found both pathologic eyes. Quantitation levels fluorescence showed significantly higher expression eyes than ones (P , 0.0001). CD40L variably inconstantly expressed cells. A strong observed eyes, whereas very low Significantly positive correlations among levels. basal conditions, negative. increased IFNg treatment hours’ exposure TNFa. These cytokines had no effect on expression. but remained negative CONCLUSIONS. Human normally express and, more inconsistently, CD40L. Flow these molecules expression, as well upregulation proinflammatory vitro. (Invest Ophthalmol Vis Sci. 2000;41:120 ‐126)
