摘要: Elucidation of the pathophysiological basis platelet disorders in murine models requires a reliable method for frequent determinations counts individual mice. Here, we present rapid, reproducible and accurate flow cytometric enumeration platelets that involves fluorescent staining whole blood with specific antibody addition known numbers beads standardization sample volume. Analysis obtained by tail bleeding indicated this sampling procedure did not activate platelets, only five microliters were required counting. Using method, followed mice infected relapsing fever spirochete Borrelia turicatae 26 days, found bacterium induces thrombocytopenia, common manifestation human fever. Therefore, can be used to follow number activation state circulating from over extended periods time is applicable wide range disorders.
