摘要: Objective: To investigate the effects of Gymnema montanum leaf extract against endoplasmic reticulum (ER) stress-induced toxicity in endothelial cells. Methods: The immortalized hybrid cell, EA.hy926 was treated with different concentrations (0-100 μg/mL) and ER stress inducer, tunicamycin. cytotoxicity assessed by MTT as well lactate dehydrogenase malondialdehyde levels were determined. markers, GRP78 CHOP analysed Western blot assay. extract-mediated activation nuclear factor erythroid 2-related 2 (Nrf2) cell-based luciferase enzyme fragment complementation assay antioxidant responsive element driven reporter phosphoproteins MAPK pathway analyzed using Bioplex system. Results: A dose-dependent cytoprotective effect observed tunicamycin-induced toxicity. significantly reduced activity It also suppressed markers dose dependently inhibited phosphorylation JNK, ERK, MEK p38 Moreover, increased expression Nrf2 its downstream targets Conclusions: attenuates increasing genes.
