摘要: Previous attempts to relate the structure and function of hepatocytic membranes have compared biochemical data fractions morphological derived from either intact tissue or fractions. The effects original homogenization aside, recoveries comparing membrane marker enzymes homogenate subsequent suggest a general conservation activity. A sterological study was undertaken estimate surface areas in tissue, homogenate, same livers then test comparability these with by calculating both recoveries. were corrected for errors due section thickness compression. average total sufrace area per 1 g liver 9.3 m2 (T), 7.8 (H), 7.4 (F); thus averaged 96% (F/H) 81% (F/T) comparisons. In fractions, differentiability criteria limited rough- smooth- surfaced membranes, as well outer inner mitochondrial membranes. rough-surfaced 101% F/H 92% F/T; those smooth-surface 89% 107% F/T. For recovery 100% obtained, whereas it amounted only 54% With respect F/H, compare enzyme obtained biochemically. extension calculations revealed satisfactory procedures fractionation; indicates, however, that shift substantial part pool unidentifiable smooth may occur on homogenization.
