摘要: The minimum size fragment of human cytomegalovirus AD169 required to initiate transformation was determined by transfection primary rat embryo cells with deletion fragments constructed digestion a cloned containing the transforming region (pCM4000) exonuclease III and S1 nuclease. results indicate that left-hand boundary sequence for must reside between 490 318 bases from HindIII site pCM4000. right-hand defined EcoRI which is 20 site. nucleotide chemical degradation technique Maxam Gilbert dideoxynucleoside triphosphate chain termination method. pCM4000 comprises 2,848 base pairs has an A X T composition 59.5%. Reading frame analysis indicated longest open reading 118 amino acids in length. Northern blot polyadenylated non-polyadenylated RNA extracted at immediate early late times after infection one 5.0-kilobase species hybridized (Jahn et al., J. Virol, press). direction transcription hybridization this transcript M13 single-stranded probes, did not detect introns.
