Effect of the DGAT1 K232A genotype of dairy cows on the milk metabolome and proteome

摘要: Diglyceride O-acyltransferase 1 (DGAT1) is the enzyme that catalyzes synthesis of triglycerides from diglycerides and acyl-coenzyme A. The DGAT1 K232A polymorphism was previously shown to have a significant influence on bovine milk production characteristics (milk yield, protein content, fat fatty acid composition). mechanism this has, however, not been elucidated. In study, metabolomics (1H-nuclear magnetic resonance) proteomics (laser chromatography-tandem mass spectrometry) were applied determine serum lipid metabolite composition globule membrane proteome samples cows with KK AA genotypes. genotype contained more stomatin, sphingomyelin, choline, carnitine, less citrate, creatine or phosphocreatine, glycerol-phosphocholine, mannose-like sugar, acetyl sugar phosphate, uridine diphosphate (UDP)-related orotic compared genotype. Based these results, we propose differences between genotypes may be related stomatin-sphingomyelin rafts as well structural (cell membrane) in epithelial cells mammary gland. conclusion, our study shows that, addition described changes triglyceride composition, differing differ their metabolome, which help further understanding effect characteristics.
