Intracellular mechanisms of aminoglycoside-induced cytotoxicity

摘要: Since introduction into clinical practice over 60 years ago, aminoglycoside antibiotics remain important drugs in the treatment of bacterial infections, cystic fibrosis and tuberculosis. However, ototoxic nephrotoxic properties these are still a major problem. Recent advances molecular biology biochemistry have begun to uncover intracellular actions aminoglycosides that lead cytotoxicity. In this review, we discuss binding targets aminoglycosides, highlighting specific aminoglycoside-binding proteins (HSP73, calreticulin CLIMP-63) their potential for triggering caspases Bcl-2 signalling cascades involved aminoglycoside-induced We also strategies reduce cytotoxicity, which necessary greater bactericidal efficacy during pharmacotherapy.
