摘要: Development of viable cell estimation method without sacrificing proliferation and functions cells cultured on regenerative biomaterials is essential for engineering. Cytotoxicity depletion resazurin are critical but often overlooked limitations that hindered applications in estimation. The present work found cytotoxicity depended concentration, concentration incubation time. A simple strategy which only allowed to incubate with during each measurement was developed eliminate negative effects resazurin. This verified by monitoring MC3T3-E1 preosteoblasts poly(d,l-lactic acid) scaffold a continuous 3D culture process up 21 days, comparing the accuracy MTT assay destructive high sensitivity commonly used engineering viability, differentiation MC3T3-E1, were treated with/without this nondestructive evaluation. showed comparable linearity standard curve characteristics growth assay. No major viability found. Our highlighted importance time designing application-specific would be helpful improving implanted medical devices as well
