摘要: CXXC finger protein 1 (CFP1) binds to unmethylated CpG motifs in DNA, is a component of the mammalian Set1 histone methyltransferase complex, and essential for zebrafish hematopoiesis. Transfection human PLB-985 myeloid cell line with short hairpin RNA directed against transcript encoding CFP1 results 80% fewer colonies compared vector control, suggesting that required survival cells. One clone, CFP1–AS1, exhibits 70% decrease levels slower doubling time due an increase proportion cells G1 G2 S phase. CFP1–AS1 exhibit 40% reduction DNA but contain normal global genomic cytosine methylation.
