摘要: The leukemic B lymphocytes from chronic lymphocytic (CLL) patients have a long survival in vivo, although ex vivo they rapidly die by apoptosis. To further investigate the mechanism of this, we studied influence bone marrow stromal cells normal subjects on apoptosis B-CLL and umbilical cord blood (UCB) lymphocytes. After 48 hours incubation medium alone, showed, respectively, 22 +/- 3% 31 5% Cocultures with reduced percentage undergoing (8 2%, P < . 0005) prevented loss bcl-2 protein expression. In contrast, slightly increased B-cell (37 6%). Direct contact between was found to be essential for inhibition cell apoptosis; indeed, separation microporous membrane spontaneous apoptosis, comparable results were obtained conditioned medium. difference behavior observed plated can explained fact that only few adhere comparison cells. adhesion is mediated beta1 beta2 integrins acting simultaneously. Contact seems play major role accumulation marrow.
