Separation and quantification of lignans in Phyllanthus species by a simple chiral densitometric method

摘要: A sensitive, selective, and robust high-performance TLC (HPTLC) method using chiral plates for qualitative quantitative analysis of phyllanthin (A), hypophyllanthin (B), niranthin (C), nirtetralin (D), the active lignans Phyllanthus species, was developed validated. The effectiveness role various stationary phases viz silica gel 60F254, HPTLC in quantitation were evaluated. precoated plate found suitable simultaneous four pharmacologically lignans. For achieving good separation, optimized mobile phase n-hexane/acetone/1,4-dioxane (9:1:0.5 by volume) used (Rf = 0.30, 0.36, 0.41, 0.48 compounds A, B, C, D, respectively). densitometric determination above carried out reflection/absorption mode at 620 nm. Optimized chromatographic conditions provide well-separated compact bands tested calibration curves linear concentration range 100 – 500 ng/band. Recoveries D 99.98, 100.51, 99.22, 98.74%, respectively. validated according to ICH guidelines. reported here is reproducible applied leaves i. e., P. amarus, maderaspatensis, urinaria, virgatus.
