摘要: Immunologic probes have been used to examine the conformation of apolipoprotein B (apo-B) as it exists within native low density lipoprotein (LDL) after lipid displacement with Triton X-100 and denaturation guanidine hydrochloride organic solvent delipidation reconstitution X-100. Antigenic expression was assayed in two systems: by using either or bovine serum albumin maintain protein solubility. Apo-B delipidated virtually identical LDL-apo-B both systems, polyclonal antisera prepared rabbits against antigen. Thus antigenic sites are preserved, although core LDL drastically alters physical properties particle. extraction reconstituted several methods, products were examined immunologically. One method yielded a product that resembled apo-B X-100, full could not be achieved. Nevertheless, promoted refolding reform partial structure judged By immunologic methods for assessing structure, is clearly evident perceptions conformational states can very different, multiple criteria need assess reconstitution.
