Inhibition of Anti-HIV MicroRNA Expression: A Mechanism for Opioid-Mediated Enhancement of HIV Infection of Monocytes

摘要: Several micro RNAs (miRNAs) have the ability to inhibit HIV replication in target cells. Thus, we investigated impact of opioids (morphine and heroin), widely abused drugs among people infected with HIV, on expression cellular anti-HIV miRNAs monocytes. We found that morphine-treated monocytes expressed lower levels than untreated In addition, morphine treatment compromised type I interferon (IFN)–induced miRNA expression. These findings paralleled observation enhanced replication. morphine-mediated actions could be antagonized by opioid receptor antagonists (naltrexone or Cys2, Tyr3, Arg5, Pen7-amide). Furthermore, vitro was confirmed vivo heroin-dependent subjects had significantly (miRNA-28, 125b, 150, 382) peripheral blood mononuclear cells healthy subjects. indicate use impairs intracellular innate mechanism(s) monocytes, contributing cell susceptibility infection.
