摘要: Spermatogonia are the male germ stem cells that continuously produce sperm for next generation. Spermatogenesis is a complicated process proceeds through mitotic phase of cell renewal and differentiation, meiotic phase, postmeiotic spermiogenesis. Full recapitulation spermatogenesis in vitro has been impossible, as generation normal spermatogonial lines without immortalization production motile from these after long-term culture have not achieved. Here we report derivation line mature medakafish testis immortalization. After 140 passages during 2 years culture, this retains stable but growth factor-dependent proliferation, diploid karyotype, phenotype gene expression pattern cells. Furthermore, show can undergo meiosis spermiogenesis to generate sperm. Therefore, ability continuous proliferation an intrinsic property medaka cells, apparently necessary derive cultures. Our findings will offer unique opportunity study recapitulate develop approaches germ-line transmission.
