Up-regulation of survivin in oral squamous cell carcinoma correlates with poor prognosis and chemoresistance.

摘要: Survivin, a key member of the inhibitor apoptosis protein family, has been reported to be capable regulating both cellular proliferation and apoptotic cell death. This is found overexpressed in many human cancers. The aim this study was evaluate prognostic significance survivin mRNA expression oral squamous carcinoma (OSCC) analyze its correlation with chemoresistance. Reverse-transcription polymerase chain reaction assay performed detect OSCC lines or tissue samples. Immunohistochemistry tissues corresponding nontumor tissues. Then between clinicopathologic features prognosis patients analyzed. Small interfering RNA technology used down-regulate gene line. Methylthiazol tetrazolium flow cytometric assays were line (HSC-3). Furthermore, effect small (siRNA) targeting on sensitivity cells chemotherapeutic agents (cisplatin 5-fluorouracil [5-FU]) determined. Results showed that levels significantly higher than those normal keratinocyte noncancerous immunostaining stronger Moreover, high correlated poorer tumor differentiation, clinical stage, presence lymph node metastasis (P < .05). Multivariate analysis status could an independent factor for (hazard ratio 2.71, 95% confidence interval 1.46-5.10 ; P = .012). In addition, siRNA-mediated down-regulation inhibit induce cells. Suvivin also enhance chemosensitivity cells, which associated enhancement. Thus, potential patients, become novel strategy chemosensitization OSCCs.
