摘要: In multiple sclerosis, myelin repair is generally insufficient despite the relative survival of oligodendrocytes within plaques and recruitment oligodendrocyte precursors. Promoting remyelination appears to be a crucial therapeutic challenge. Using newly developed enzymatic index myelination, we screened different neurotrophic factors for their ability enhance myelination. Neurotrophins [NGF, neurotrophin-3 (NT-3), NT-4/5, BDNF], glial cell line-derived factor (GDNF)-related (GDNF, neurturin), growth such as PDGF-AA, FGF-2, insulin did not increase myelinogenesis. contrast, among belonging CNTF family, CNTF, leukemia inhibitory factor, cardiotrophin-1, oncostatin M induced strong promyelinating effect. We provide evidence that acts on by favoring final maturation, this effect mediated through 130 kDa glycoprotein receptor common family transduced Janus kinase pathway. Our results demonstrate novel role raise possibility these might interest promote in sclerosis.
