Evaluation of platelet-rich plasma from diabetic donors shows increased platelet vascular endothelial growth factor release

摘要: Background: Platelet-rich plasma (PRP) contains pro-angiogenic growth factors including vascular endothelial factor (VEGF). Angiogenesis is a necessary component of wound healing in instances diabetic foot ulcers (DFU). PRP composition varies depending on methods and donor health status. Our group has developed an improved protocol for diabetes treatment. The aims this study were to examine the levels VEGF these patient populations with without diabetes. Methods: was prepared using 24 mL whole blood from 13 10 non-diabetic patients registered at Klinik Hayandra. Whole sodium citrate tubes centrifuged 1,000 rpm 5 minutes followed by separation. Plasma samples 3,000 minutes. Upper platelet-poor layers discarded, leaving concentrated platelet containing (PRP). Concentrated mixed, aliquoted, stored −86 °C, pooled count, VEGF, total protein analyses. Platelet counting also performed fresh measure changes following preparation. Results: Diabetic donors had higher counts than donors, but difference not statistically significant. An average increase more 250% number after preparation our method noted both groups. Freezing-thawing °C lysed 90% platelets regardless lower mean concentrations. Lysed released those donors. Conclusions: content making autologous application promising treatment DFU. However, should be investigated another appropriate clinical trial.
