Upregulation of APP, ADAM10 and ADAM17 in the Denervated Mouse Dentate Gyrus

摘要: The disintegrin and metalloproteinases ADAM10 ADAM17 are regarded as the most important α-secretases involved in physiological processing of amyloid precursor protein (APP) brain. Since it has been suggested that APP by could be reorganization brain following injury, we studied mRNA expression two Adam10 Adam17, s-secretase Bace1, App-gene family (App, Aplp1, Aplp2) dentate gyrus mouse entorhinal denervation. Using laser microdissection, tissue was harvested from outer molecular layer granule cell denervated gyrus. Expression levels candidate genes were assessed using Affymetrix GeneChip Mouse Gene 1.0 ST arrays reverse transcription-quantitative PCR, revealing an upregulation Adam17 App layer. Immunolabeling for or combination with markers astro- microglia revealed increased labeling associated reactive astrocytes but not microglia. Collectively, these data show denervation affects level its α-secretases. This suggests APP-processing shifted towards non-amyloidogenic pathway areas and, thus, formation neuroprotective cleavage products, such APPsα.
