Detection of monoclonal antibodies specific for carbohydrate epitopes using periodate oxidation

摘要: A method is described for determining whether particular monoclonal antibodies are specific carbohydrate or non-carbohydrate antigenic determinants. In a model system consisting of the Lewis human blood group determinant attached to either protein lipid, mild periodate oxidation destroyed without altering lipid epitopes. The technique was readily applied antigens bound plastic wells ELISA, nitrocellulose sheets Western blots, and thin layer chromatography (TLC) plates TLC immunostaining. Mild can prove useful during early stages hybridoma screening in order select against anti-carbohydrate antibodies.
