Cytokeratin 8 and 19 as antigens recognized by adenocarcinoma-reactive human monoclonal antibody AE6F4

摘要: The human monoclonal antibody (MAb) AE6F4 is secreted by a human-human hybridoma line established from the in vitro immunization of normal peripheral blood lymphocytes with lung adenocarcinoma cell line, A549. This MAb strongly reactive to cancer tissues. In previous study, antigens recognized were purified A549 cells and identified as /4-3-3 protein 31 kDa cytosolic phospholipase A2 (cP LA2)' also binds two kinds (53 40 kDa), which are not related or cPLA2, breast MCF-7. We 38 antigen, degradation product 53 antigen MCF-7 using ion-exchange hydroxyapatite column chromatography. Two partial amino acid sequences showed 95-100% homology cytokeratin 8 (CK8). Two-dimensional gel electrophoresis immunoblot analysis intermediate filament fraction separated demonstrated that CK8 CK19, respectively. Antigenic determinants on CK19 resistant sodium periodate treatment, although antigenic determinant (14-3-3 and( or) cP LA2) was, sensitive this treatment. These results suggest reacts both carbohydrate peptide determinants. [Hum Antibod 1997; 8: 195-202]
