摘要: Nicotine stimulates the release of neurotransmitters in CNS. One locus its action is directly on nerve terminal, through presynaptic nicotinic acetylcholine receptors. The stimulation dopamine from striatal terminals has been particularly widely studied. acts a dose-dependent manner (EC50=4µM) to elicit Ca2+-dependent release. pharmacological profile this favours ganglionic type receptor, but molecular biological techniques have revealed several subtypes receptors specificity nicotine-evoked transmitter consistent with receptor class identified by high affinity [3H]nicotine binding. This correlation supported loss such binding sites following degeneration after lesion experiments or degenerative diseases, and subcellular fractionation expermients indicate that proportion are associated isolated terminals. Thus may constitute major target nicotine brain.
