摘要: The ability to introduce any chemical probe endogenous target protein in its native environment, that is cells and vivo, anticipated provide various new exciting tools for biological biomedical research. Although still at the prototype stage, ligand-directed tosyl (LDT) chemistry a novel type of affinity labeling technique we developed such dream. This allows modifying proteins by probes with high specificity settings ranging from vitro (in test tubes) living vivo. Since first report, list are successfully labeled LDT has been increasing. A growing number studies have demonstrated utility create semisynthetic directly cellular contexts. situ generated applicable types analysis imaging intracellular processes. In this review, summarize basic properties applications toward engineering systems. Current limitations future challenges area also described.
