摘要: Our laboratories have previously reported on the experimental infection of cattle with Mycobacterium avium subsp paratuberculosis (M. paratuberculosis) using an intratonsillar model. In addition, we recently developed a partial protein array representing 92 M. coding sequences. These combined tools enabled unique look at temporal analysis antigens within native host. The primary objective this study was to identify detected by early during infection. A secondary evaluate humoral immune response in initial year Sera from two experimentally infected cattle, taken pre-inoculation and day 70, 194 321 post infection, identified dynamic antibody reactivity among some showing increased over time others declining levels same period. specific protein, encoded MAP0862, strongly initially, but became weaker time. most reactive putative surface antigen MAP1087. second MAP1204, implicated virulence, also 70 both cattle. Subsequent experiments showed that these proteins were sera 5 9 naturally subclinical stage Johne's disease. Collectively results demonstrate are as days after exposure. data further suggest least may be useful diagnosis infections. Finally, construction use pilot has led novel approach for discovery antigens.
