Killing of Gram-Negative Bacteria with Normal Human Serum and Normal Bovine Serum: Use of Lysozyme and Complement Proteins in the Death of Salmonella Strains O48

摘要: Serum is an environment in which bacterial cells should not exist. The serum complement system provides innate defense against microbial infections. It consists of at least 35 proteins, mostly pre-activated enzymatic forms. activation achieved through three major pathways: the classical, alternative, and lectin. Lysozyme, widely present body fluids, catalyzes hydrolysis β 1,4 linkage between N-acetyloglucosamine N-acetylmuramic acid cell wall cooperates with bactericidal action serum. In this study, ten strains serotype O48 Salmonella, mainly associated warm-blooded vertebrates clinically important causing diarrhea infants children, were tested. results demonstrated that most efficient killing Salmonella occurred when all components normal bovine (NBS) human (NHS) cooperated. To prove role lysozyme activity serum, method adsorption onto bentonite (montmorillonite, MMT) was used. order to investigate structural transitions accompanying components, we applied X-ray diffraction methods. investigation suggested apart from lysozyme, other proteins (as, e.g., C3 protein or IgG immunoglobulin) adsorbed on MMT particles. also shown Ca2+ cations can be bentonite. This may explain different sensitivities serovars belonging same NBS NHS devoid lysozyme.
