Inhibition of cytokinesis in Friend leukemia cells by membrane mobility agents.

摘要: Treatment of a line Friend leukemia cells with dispersion the membrane mobility agent, A2C, yields that undergo successive nuclear divisions without cytokinesis, resulting eventually in as many 30 nuclei. Neither DNA replication rate nor generation time is different after treatment; addition, multiple nuclei divide synchronously, and chromosome number corresponds to cell. Inhibition cytokinesis not observed if are washed reagent-free medium within 1 hr treatment, but washing delayed for 24 hr. Membrane agent loaded fluorescent probe, Flomol F20C, leads membrane; fluorescence disappears from change hr, Some stages overall development resemble those seen cytochalasin B inhibition although mechanisms may well be promoted by agent. The A2C provides potentially interesting means studying regulation differentiation.
