摘要: The MSC16 cucumber (Cucumis sativus L.) mutant with lower activity of mitochondrial Complex I was used to study the influence metabolism on whole cell energy and redox state. Mutant plants had content adenylates NADP(H) whereas NAD(H) pool similar as in wild type. Subcellular compartmentation pyridine nucleotides were studied using method rapid fractionation protoplasts. data obtained demonstrate that dysfunction respiratory chain decreased chloroplastic ATP pool. No differences pools subcellular fractions mutated observed; however, cytosolic fraction highly reduced more oxidized MSC16, compared WTc. protoplasts greatly reduced, extrachloroplastic much oxidized, than WTc protoplast. Changes distribution changes found tobacco (Nicotiana sylvestris) CMS II mutant. In contrast cucumber, higher pronounced leaf tissue collected after darkness middle photoperiod. Results protoplast fractionating showed increase adenylate mainly due a level extrachloroplast fraction. Both mutations have negative effect plant growth through perturbation chloroplast/mitochondrial interactions.
