Syntenin, a PDZ protein that binds syndecan cytoplasmic domains

摘要: The syndecans are transmembrane proteoglycans that place structurally heterogeneous heparan sulfate chains at the cell surface and a highly conserved polypeptide in cytoplasm. Their versatile moieties support various processes of molecular recognition, signaling, trafficking. Here we report identification protein binds to cytoplasmic domains yeast two-hybrid screens, plasmon resonance experiments, ligand-overlay assays. This protein, syntenin, contains tandem repeat PDZ reacts with FYA C-terminal amino acid sequence syndecans. Recombinant enhanced green fluorescent (eGFP)–syntenin fusion proteins decorate plasmamembrane intracellular vesicles, where they colocalize cosegregate Cells overexpress eGFP–syntenin show numerous extensions, suggesting effects syntenin on cytoskeleton–membrane organization. We propose may function as an adaptor couples cytoskeletal or cytosolic downstream signal-effectors.
