摘要: This study was undertaken to investigate the phenotypic and functional status of T lymphocytes human fetuses from early second- third-trimester. Cord blood samples were obtained 19 healthy (gestation weeks: 18-36), by cordocentesis, 16 term newborns weeks 37-42). Maternal unrelated male also taken as controls. Percentage in fetal white cells 79.3%, reducing 40% birth, much higher than that adults. mononuclear (CBMC), prepared density gradient centrifugation followed lysis erythrocytes, stained using PE- or FITC-labelled monoclonal Abs analysed flow cytometry. The frequencies CD3+ (40.1%) neonatal (42.4%) CBMC significantly lower men (59.6%) pregnant women (53.6%). Proportions CD8+ (9.5%), gammadelta-T (0.5%) NK (4.8%) neonates (except cells) A negative linear correlation (r = -0.609) between ratio CD4+/CD8+ gestation age could be established. Fetal showed vigorous spontaneous proliferation but failed respond mitogen (PHA) allogeneic stimulation vitro. unable produce IL-2, IL-4 IFN-gamma, spontaneously secreted IL-10, IL-6 TNF-alphain Stimulation with PHA up-regulated production TNF-alpha substantially.
