摘要: Abstract Chancroid is a sexually transmitted genital ulcer disease caused by Haemophilus ducreyi. Previously, we developed diagnostic DNA probes for H. ducreyi (L. M. Parsons, Shayegani, A. L. Waring, and Bopp, J. Clin. Microbiol. 27:1441-1445, 1989). In the present study, sequencing of one revealed two adjacent open reading frames (ORFs). These ORFs encoded proteins show significant homology with groE genes GroES GroEL heat shock from several bacterial pathogens conserved eukaryotic 60-kDa proteins. The first ORF (groES) preceded sequences similar to those Escherichia coli consensus promoters 288 nucleotides long capable encoding protein 10.3 kDa. second (groEL) 1,641 57.8 Northern (RNA blot) analysis demonstrated presence high level mRNA in exponential-phase grown hemin broth at organism's optimal growth temperature (33 degrees C), increased levels seen following shock. Heat also thermostability organisms, since stressed cells were more resistant lethal effects rapid chilling. Electrophoretic immunoblots that predominant produced was heat-inducible, immunoreactive approximately 60 kDa (GroEL). Also, expressed inducible E. coli. This report describing cloning, sequencing, expression protein-encoding genes.
