摘要: The present study sought to determine whether in vitro maturation (IVM) of pig oocytes a medium supplemented with insulin growth factor-I (IGF-I) and subsequent vitrification or without reduced glutathione (GSH) affect their quality developmental competence, the expression genes involved antioxidant, apoptotic stress responses. In Experiment 1, cumulus-oocyte complexes were matured absence presence IGF-I (100 ng·mL-1) then vitrified-warmed 2 mM GSH. Maturation rate was evaluated before vitrification, oocyte viability, DNA fragmentation relative transcript abundances BCL-2-associated X protein (BAX), BCL2-like1 (BCL2L1), heat shock 70 (HSPA1A), peroxidase 1 (GPX1) superoxide dismutase (SOD1) assessed fresh oocytes. 2, fertilized competence determined. Whereas addition had no effect on maturation, it caused an increase survival This accompanied by concomitant augment abundance HSPA1A decrease BAX. Furthermore, GSH vitrification-warming media increased rates at post-warming. Likewise, action GPX1 BAX transcript. Blastocyst did not differ from counterparts when combined. conclusion, supplementing 100 ng·mL-1 solutions improves cryotolerance IVM oocytes, through mechanism that involves BAX, expression.
