Insulin-like growth factor I promotes oocyte maturation through increasing the expression and phosphorylation of epidermal growth factor receptor in the zebrafish ovary.

摘要: The resumption of oocyte meiosis is a critical step for the progression development, which requires an intimate collaboration variety hormones and growth factors. Insulin-like factor I (IGF-I) epidermal (EGF) family are well recognized to promote maturation. However, mechanism by they coordinate this process remains unknown. present study demonstrated that IGF-I can increase egfr mRNA protein levels in follicle cell culture or intact follicles. This stimulation be significantly inhibited IGF-IR specific inhibitor, NVP-ADW742. inhibitors against phosphatidylinositol-3-kinase (PI3K), phosphoinositide-dependent kinase 1 (PDK1) Akt also dramatically abolished IGF-I-induced expression, suggesting classical PI3K/Akt pathway mediated action regulation. We further found not only was level Egfr increased, but phosphorylation enhanced IGF-I. Unlike egfr, failed stimulate expression Egf-like ligands whereas decreased protein-tyrosine phosphatase, receptor type, kappa (ptprk), tyrosine phosphatase. maturation assay confirmed initiates regulation through its cognate cells. Taken together, promoted maturation, part at least, ligands/Egfr pathway. sheds light on cross-talk between two important factors zebrafish ovary underlying induction
