A His2AvDGFP fusion gene complements a lethal His2AvD mutant allele and provides an in vivo marker for Drosophila chromosome behavior.

摘要: We have generated Drosophila melanogaster lines carrying a modified genomic fragment which encodes the D. variant H2A.F/Z class histone, His2AvD, fused to green fluorescent protein (GFP) of jellyfish Aequorea victoria. show here that fusion consists functional GFP and histone His2AvD. The His2AvD portion gene was shown be by rescue mutant lethality. Fluorescence in vivo observed embryonic cleavage stage interphase nuclei on chromosomes as early cycle 9, correlating with activation transcription. Unlike transcription factors, His2AvDGFP remained transcriptionally inactive throughout mitosis. Subsequently, fluorescence at all stages larval development adult somatic tissues, consistent distribution immunohistochemical staining. This acts an excellent marker for chromosome behavior and, given ability prevent null-mutant lethality, without disrupting normal cellular functions. very high level conservation family histones suggests equivalent construct should function equally well wide range organisms.
