BODIPY(®) FL histamine as a new modality for quantitative detection of histamine receptor upregulation upon IgE sensitization in murine bone marrow-derived mast cells.

摘要: Flow cytometry is one of the most widely used methods for qualitative and quantitative analysis cell surface expressed proteins by making use fluorescent specific antibodies. Lacking an antibody validated flow cytometry, alternative approach labeling receptors fluorescently tagged ligands. In this study, histamine H4 receptor transfected Chinese hamster ovary cells murine bone marrow-derived mast (mBMMCs) were selected studying possibility staining individual using BODIPY(®) FL selective antagonists. cytometric measurements supporting calculations showed that BODIPY suitable tool quantitating receptors. The binding, competitive inhibition ligand characterized, which in good agreement with a semi-empirical model constructed from fundamental protein-binding relationships. Using method it was shown first time even though mature mBMMCs express H2R H4R to same extent, immunoglobulin E sensitization results upregulation only, while expression remains unchanged.
